Diffusion across cell membranes pt. 1- Osmosis

Experimental Set-up

Equipment and Materials

1.  OsmoDiffusion Chamber w/ wooden clamp.
2. stir plate
3. small stir bar
4. 30 mL syringe w/ tubing
5. 1mL flow rate pipet w/ cork
6. Sugar (Mannitol or Glucose) solutions in phosphate buffer
7. Control, phosphate buffer

Add sugar solution to cell side

Add phosphate buffer to environment side

 

 

 

<Add stir bar to the Cell side.



<Membrane (100 MWCO) is sandwiched between the two sides of the chamber.

Do Not damage it when adding solutions to the chamber

Reading the pipet & calculating flow.

1. Mark the pipet at the start.

2. Each large increment = 0.1 mL, Each small increment = 0.01 mL

3.  If it takes 3 min for the solution to move from 0.11 mL to 0.26 mL,

then the flow or osmotic rate = 0.15 mL/3 min, or 0.05 mL/min
